Allicin — The Bioactive Organosulfur Compound in Garlic

Allicin is the primary biologically active organosulfur compound produced when fresh garlic is crushed or chopped. It exhibits potent antioxidant, anti-inflammatory, and antimicrobial properties, contributing to cardiovascular health and potentially extending healthspan by mitigating age-related oxidative stress.

Mechanism of Action

Allicin is formed from the precursor alliin by the enzyme alliinase upon tissue damage in garlic. It rapidly decomposes into various sulfur-containing compounds, such as diallyl disulfide (DADS) and diallyl trisulfide (DATS). These compounds act as hydrogen sulfide (H2S) donors, which activate endothelial nitric oxide synthase (eNOS) to promote vasodilation. Furthermore, allicin and its derivatives upregulate the Nrf2/HO-1 antioxidant pathway, enhancing the expression of endogenous antioxidant enzymes and suppressing NF-κB-mediated inflammatory signaling.

Human Trial Evidence

Numerous human clinical trials have investigated garlic supplements, though specific trials isolating pure allicin are less common due to its instability. Meta-analyses of randomized controlled trials show that garlic preparations significantly reduce blood pressure in hypertensive individuals and modestly lower total cholesterol and LDL levels. However, direct human longevity trials are lacking, and evidence primarily supports its role in reducing cardiovascular disease risk factors.

Dosing Protocol

Consuming 1–2 cloves (approx. 3–6 grams) of raw, crushed garlic daily yields about 3–5 mg of allicin. For supplements, aged garlic extract (AGE) or allicin-yielding powders are used, typically dosed at 600–1,200 mg daily. It is best consumed with food to minimize gastrointestinal irritation.

Safety & Contraindications

Generally recognized as safe in culinary amounts. High doses or raw garlic consumption can cause gastrointestinal distress, heartburn, and body odor. Due to its antiplatelet properties, high doses should be used with caution in individuals taking anticoagulant medications (e.g., warfarin) or prior to surgery due to an increased risk of bleeding.
