Bromantane — The Atypical Actoprotector
Bromantane is a synthetic adamantane derivative that functions as an actoprotector, combining mild psychostimulant and anxiolytic properties. While it is not a direct longevity intervention, its ability to enhance physical and mental endurance without the typical depletion associated with classical stimulants makes it of interest for mitigating age-related fatigue and cognitive decline.
Mechanism of Action
Bromantane is classified as an actoprotector and atypical stimulant. Its primary mechanism involves the upregulation of tyrosine hydroxylase and aromatic L-amino acid decarboxylase, the rate-limiting enzymes in dopamine synthesis, thereby increasing dopamine levels in the brain without directly releasing it or significantly blocking its reuptake. It also exerts anxiolytic effects by modulating the GABAergic system, strengthening GABA-mediated neurotransmission. Additionally, it has been shown to influence serotonin reuptake and exhibit mild immunomodulatory properties.
Human Trial Evidence
Bromantane (under the brand name Ladasten) has been evaluated in Russian clinical trials for the treatment of neurasthenia and asthenic disorders. A 2009 randomised, placebo-controlled study by Neznamov et al. reported significant reductions in asthenic symptoms and anxiety compared to placebo. However, there are no published human longevity trials, and its use remains largely confined to specific regional medical practices and off-label cognitive enhancement.
Dosing Protocol
Typical dosing ranges from 50–100 mg per day, taken orally. Due to its long half-life and stimulating properties, it is generally administered in the morning or early afternoon to avoid sleep disruption. As an atypical stimulant with potential for misuse, caution is advised, and it is not approved by the FDA or EMA for clinical use.
Safety & Contraindications
Bromantane is generally considered to have a lower risk of dependence and cardiovascular strain compared to classical psychostimulants like amphetamines. However, potential adverse effects include insomnia, irritability, and gastrointestinal distress. Due to its dopaminergic activity, it carries a risk of abuse and should be used with caution, particularly in individuals with a history of substance use disorders or severe psychiatric conditions. It is banned by the World Anti-Doping Agency (WADA) in competitive sports.
