Centrophenoxine is a cholinergic nootropic and anti-aging compound best known for its ability to clear lipofuscin, a cellular waste product that accumulates with age. While it has demonstrated lifespan extension in mice and neuroprotective effects in older adults, its longevity benefits in humans remain unproven.

Mechanism of Action

Centrophenoxine (meclofenoxate) is an ester of dimethylethanolamine (DMAE) and 4-chlorophenoxyacetic acid (pCPA). It acts as a cholinergic nootropic by increasing the availability of choline and acetylcholine in the brain. In the context of aging, its primary mechanism is the reduction of lipofuscin, an age-related pigment that accumulates in post-mitotic cells like neurons. Human Trial Evidence

Human trials are limited and mostly dated. A few older double-blind clinical trials in elderly patients with dementia or cognitive decline reported improvements in memory and cognitive function. However, there are no published human longevity trials demonstrating lifespan extension; evidence for longevity is restricted to animal models, where it has been shown to increase the lifespan of mice.

Dosing Protocol

250–1,000 mg/day is the most commonly used range, often divided into two doses. It is typically taken with food to improve absorption. Due to its stimulatory nature, it is best taken in the morning or early afternoon to avoid insomnia. As an unapproved drug in the US, it is sold as a dietary supplement, but it is a prescription drug in some European countries and Japan.
