Chromium picolinate is a highly bioavailable form of the essential trace mineral chromium, widely studied for its role in carbohydrate and lipid metabolism. While primarily known for its potential to enhance insulin sensitivity and support glycemic control, its relevance to longevity stems from the critical role of glucose homeostasis and insulin signaling in aging and metabolic health.
Mechanism of Action
Chromium is an essential trace mineral that appears to enhance the action of insulin. The precise molecular mechanism is not fully elucidated, but it is thought to involve the formation of a low-molecular-weight chromium-binding substance (chromodulin) that binds to and activates the insulin receptor kinase. This activation amplifies the insulin signaling cascade, leading to increased translocation of GLUT4 transporters to the cell membrane and enhanced cellular glucose uptake. It may also transiently upregulate AMPK, further supporting glucose disposal and lipid metabolism.
Human Trial Evidence
Numerous human trials have investigated chromium picolinate for glycemic control and insulin resistance, primarily in populations with type 2 diabetes or metabolic syndrome. Results are mixed, with some studies showing modest improvements in fasting glucose, insulin sensitivity, and HbA1c, while others show no significant benefit. No published human longevity trials exist.
Dosing Protocol
200–1,000 mcg/day is the most studied range in humans for metabolic support. Typically taken once or twice daily with meals to support glucose metabolism. Doses above 1,000 mcg/day are generally not recommended due to theoretical risks of renal or hepatic toxicity.
Safety & Contraindications
