Cranberry Extract — Proanthocyanidin-Rich Polyphenol
Cranberry extract is a concentrated source of A-type proanthocyanidins and other polyphenols known for their potent antioxidant and anti-inflammatory properties. In longevity research, it has demonstrated the ability to extend lifespan and improve healthspan in model organisms by modulating stress resistance and proteostasis pathways.
Mechanism of Action
The longevity effects of cranberry extract are primarily mediated by its high concentration of proanthocyanidins. In model organisms like C. elegans, it extends lifespan by activating the DAF-16/FOXO transcription factor and the OSR-1 pathway, which enhance cellular stress resistance. It also improves proteostasis by upregulating heat shock factor 1 (HSF-1), thereby reducing amyloid toxicity. Additionally, its potent antioxidant capacity neutralizes reactive oxygen species (ROS) and modulates the p38 MAPK pathway to promote innate immunity.

Dosing Protocol
For general health and urinary tract support, typical doses range from 300–500 mg of standardized cranberry extract daily, often standardized to contain 36 mg of proanthocyanidins (PACs). Dose-finding for longevity is unestablished in humans.
Safety & Contraindications
Generally well-tolerated at standard doses. High doses may cause gastrointestinal upset or increase the risk of calcium oxalate kidney stones in susceptible individuals due to its oxalate content. It may interact with blood thinners like warfarin, potentially increasing bleeding risk.
