Dasatinib — The Tyrosine Kinase Senolytic
Dasatinib is a tyrosine kinase inhibitor originally developed as a leukemia treatment that has emerged as a potent senolytic agent. When combined with quercetin, it selectively clears senescent cells, reducing inflammation and potentially ameliorating age-related tissue dysfunction.
Mechanism of Action
Dasatinib functions primarily as a broad-spectrum tyrosine kinase inhibitor, targeting BCR-ABL, SRC family kinases, c-KIT, and PDGFR. In the context of longevity, it acts as a senolytic by transiently disabling Senescent Cell Anti-Apoptotic Pathways (SCAPs), specifically targeting the ephrin-dependent (EFNB1/3) and PI3K/AKT survival networks. By inhibiting these pathways, dasatinib induces apoptosis in specific types of senescent cells, such as senescent preadipocytes and endothelial cells, thereby reducing the secretion of the senescence-associated secretory phenotype (SASP).
Human Trial Evidence
Early-stage human trials have explored the senolytic combination of dasatinib and quercetin (D+Q). A 2019 pilot study in patients with idiopathic pulmonary fibrosis (IPF) demonstrated that D+Q treatment improved physical function, though it did not significantly alter pulmonary function. Subsequent phase I/II trials have investigated its safety and efficacy in conditions like diabetic kidney disease and Alzheimer's disease, showing preliminary evidence of senescent cell clearance in humans.
Dosing Protocol
In senolytic protocols, dasatinib is typically used intermittently rather than continuously to minimize side effects, often dosed at 50–100 mg alongside 500–1000 mg of quercetin for 2–3 consecutive days per month. As a prescription-only chemotherapy drug, its use for longevity purposes is strictly off-label and requires medical supervision.
Safety & Contraindications
Dasatinib is a potent prescription medication with significant potential adverse effects, including myelosuppression, fluid retention (such as pleural effusion), bleeding events, and gastrointestinal toxicity. It is contraindicated in pregnancy and in individuals with active bleeding disorders. Due to its immunosuppressive and off-target effects, continuous use is avoided in longevity protocols, and it should not be used without rigorous clinical oversight.
