FOXO4-DRI Peptide — Targeted Senolytic
FOXO4-DRI is a synthetic retro-inverso peptide designed to selectively induce apoptosis in senescent cells. By disrupting the interaction between FOXO4 and p53, it represents a highly targeted experimental approach to clearing senescent cells and restoring tissue homeostasis in preclinical models of aging.
Mechanism of Action
In senescent cells, the transcription factor FOXO4 is upregulated and binds to p53, sequestering it in the nucleus and preventing it from inducing apoptosis. FOXO4-DRI is a D-retro-inverso peptide that competitively inhibits this FOXO4-p53 interaction. Upon disruption, p53 is released and translocates to the mitochondria, triggering caspase-dependent apoptosis specifically in senescent cells. Because healthy cells express lower levels of FOXO4 and do not rely on this interaction for survival, the peptide acts as a highly selective senolytic agent.
