Huperzine A is a naturally occurring alkaloid extracted from the Chinese club moss Huperzia serrata. It is primarily known for its cognitive-enhancing properties and is investigated in longevity medicine for its potential to protect against age-related neurodegeneration and cognitive decline.
Mechanism of Action
Huperzine A acts as a highly specific, reversible inhibitor of acetylcholinesterase (AChE), preventing the breakdown of acetylcholine and increasing its synaptic availability. Beyond AChE inhibition, it exhibits neuroprotective effects by reducing reactive oxygen species (ROS) generation and attenuating neuroinflammation. It has been shown to upregulate the cholinergic anti-inflammatory pathway, potentially inhibiting immunosenescence and reducing the transcription of pro-inflammatory cytokines like TNF-α.

Multiple randomized clinical trials have investigated Huperzine A in patients with Alzheimer's disease and vascular dementia, generally reporting significant improvements in cognitive function. Dosing Protocol
In clinical trials for cognitive decline, doses typically range from 200 to 400 mcg per day, often divided into two doses. For general cognitive support, supplements usually provide 50 to 200 mcg per day. It is often cycled to prevent tolerance, though clinical consensus on cycling is unestablished.
Safety & Contraindications
Generally well-tolerated at standard doses, but can cause cholinergic adverse effects including nausea, diarrhea, sweating, and muscle twitching. It is contraindicated in individuals with bradycardia, epilepsy, peptic ulcer disease, or asthma. It should not be combined with other acetylcholinesterase inhibitors or cholinergic drugs.
