Hyaluronic acid is a ubiquitous glycosaminoglycan that maintains tissue hydration, joint lubrication, and extracellular matrix integrity. While topical applications are common, oral supplementation has emerged as a systemic approach to restore declining hyaluronic acid levels associated with aging, demonstrating benefits for skin moisture and joint health.
Mechanism of Action
Orally ingested high-molecular-weight hyaluronic acid is partially degraded by gut microbiota into smaller oligosaccharides, which are absorbed into the bloodstream and distributed to peripheral tissues including the skin and joints. In the skin, it binds water to maintain extracellular matrix volume, stimulates fibroblast proliferation, and upregulates endogenous hyaluronic acid synthesis. It also interacts with CD44 and TLR4 receptors, modulating inflammatory pathways and promoting cellular repair mechanisms.
Human Trial Evidence
Multiple randomized, double-blind, placebo-controlled trials have demonstrated the efficacy of oral hyaluronic acid. Studies show that daily supplementation (120–240 mg) over 8 to 12 weeks significantly improves skin hydration, elasticity, and reduces wrinkle depth. Additional clinical evidence supports its use in reducing joint pain and improving function in patients with mild to moderate osteoarthritis.
Dosing Protocol
120–240 mg/day is the most studied and effective range in human clinical trials for both skin and joint benefits. It is typically taken once daily, and consistent use for at least 8 to 12 weeks is required to observe significant improvements.
Safety & Contraindications
Oral hyaluronic acid is widely considered safe and well-tolerated, with a very low incidence of adverse effects. Mild gastrointestinal discomfort is rarely reported. There are no known significant drug interactions, though individuals with a history of cancer should consult a physician, as hyaluronic acid metabolism is altered in some malignancies.
