Idebenone is a synthetic, short-chain analogue of Coenzyme Q10 designed to bypass complex I of the mitochondrial electron transport chain. It acts as a potent mitochondrial electron carrier and antioxidant, showing potential in protecting against cellular oxidative stress and mitochondrial dysfunction associated with aging.
Mechanism of Action
Idebenone functions as an electron carrier within the mitochondrial electron transport chain, specifically transferring electrons directly to complex III. This bypasses complex I, allowing for continued ATP production even when complex I is impaired. Unlike endogenous CoQ10, its shorter lipophilic tail prevents it from acting as a pro-oxidant under hypoxic conditions. It also upregulates ROS-detoxifying enzymes, thereby reducing oxidative damage and protecting mitochondrial integrity.
Human Trial Evidence
Idebenone has been studied in human clinical trials primarily for neurodegenerative and mitochondrial disorders, such as Friedreich's ataxia, Duchenne muscular dystrophy, and Leber's hereditary optic neuropathy (LHON). No published human longevity trials. Animal/in-vitro evidence only.
Dosing Protocol
For mitochondrial disorders like LHON, standard dosing is typically 900 mg/day, divided into three 300 mg doses taken with meals to enhance absorption. As an off-label longevity or cognitive supplement, doses often range from 150 to 300 mg/day. It is available by prescription in some countries for specific indications.
Safety & Contraindications
Generally well-tolerated at standard doses. Mild adverse effects can include gastrointestinal upset, nausea, diarrhea, and transient neurological complaints. Long-term safety for general anti-aging use is not fully established.
