Klotho is a naturally occurring protein that acts as a powerful aging suppressor, primarily by regulating mineral metabolism and suppressing insulin/IGF-1 signaling. Discovered when its mutation caused rapid aging in mice, it is now a major target in longevity research for its potential to protect against age-related cardiovascular and neurodegenerative diseases.
Mechanism of Action
Klotho is a transmembrane protein whose extracellular domain is cleaved and shed into circulation as a soluble hormone. It functions as an obligate co-receptor for Fibroblast Growth Factor 23 (FGF23), regulating phosphate and vitamin D homeostasis. Independent of FGF23, soluble Klotho suppresses insulin/IGF-1 signaling, inhibits the Wnt signaling pathway, and reduces oxidative stress. These pleiotropic effects protect against vascular calcification, preserve stem cell pools, and mitigate cellular senescence.
Human Trial Evidence
No published human longevity trials. Animal/in-vitro evidence only. Observational studies in humans correlate higher circulating Klotho levels with lower risks of cardiovascular disease, cognitive decline, and all-cause mortality, but interventional trials are lacking.
