L-theanine — The Cognitive Calming Amino Acid
L-theanine is a non-proteinogenic amino acid found primarily in green tea that promotes relaxation without drowsiness by modulating neurotransmitter balance. While its direct impact on human lifespan is unproven, it is highly valued in longevity protocols for mitigating chronic psychological stress, improving sleep quality, and supporting cognitive health—all critical factors for healthspan.
Mechanism of Action
L-theanine is a structural analogue of glutamate and glutamine that crosses the blood-brain barrier. It acts as a competitive antagonist at various glutamate receptors (including AMPA, kainate, and NMDA), thereby dampening excitatory glutamatergic neurotransmission. Simultaneously, it increases the expression of GABA, serotonin, and dopamine in specific brain regions, promoting alpha-band oscillatory brain activity associated with relaxed alertness. In preclinical models (C. elegans), L-theanine extends lifespan by activating stress resistance pathways, including DAF-16/FOXO and modulating mitochondrial dynamics and autophagy, though these longevity mechanisms remain uncharacterised in humans.
Human Trial Evidence
Multiple human trials demonstrate that L-theanine reduces psychological and physiological stress responses and improves cognitive function, particularly attention and working memory. A 4-week randomized controlled trial (Hidese et al., 2019) showed improvements in stress-related symptoms and verbal fluency at 200 mg/day. No published human longevity trials exist; lifespan extension data is limited to C. elegans models.
Dosing Protocol
200–400 mg/day is the most studied range in humans for cognitive and stress-reduction benefits. It is typically taken in single doses of 100–200 mg, often paired with caffeine (e.g., 100 mg L-theanine with 50 mg caffeine) to enhance focus while mitigating jitteriness. It can be taken with or without food.
Safety & Contraindications
L-theanine is generally recognized as safe (GRAS) with a very favorable safety profile and no severe adverse effects reported at standard doses (up to 400 mg/day). It may mildly lower blood pressure, so caution is advised for individuals on antihypertensive medications to avoid hypotension. It may also interact with stimulant medications by blunting their effects or with sedatives by enhancing them.
