Lion's mane (Hericium erinaceus) is a medicinal mushroom renowned for its neuroprotective and cognitive-enhancing properties. It contains bioactive compounds that stimulate the synthesis of nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF), making it a compelling candidate for mitigating age-related cognitive decline.
Mechanism of Action
The primary active constituents of Lion's mane are hericenones (found in the fruiting body) and erinacines (found in the mycelium). These low-molecular-weight compounds cross the blood-brain barrier and stimulate the expression of NGF and BDNF in astrocytes and neurons. This neurotrophic upregulation promotes neurite outgrowth, enhances synaptic plasticity, and protects against endoplasmic reticulum stress-induced neurotoxicity. Additionally, its polysaccharides exhibit immunomodulatory and antioxidant effects, reducing neuroinflammation via the suppression of pro-inflammatory cytokines like TNF-α and IL-6.
Human Trial Evidence
Human evidence is promising but limited to small-scale trials. A notable 2009 double-blind, placebo-controlled trial (Phytotherapy Research) in older adults with mild cognitive impairment demonstrated significant improvements in cognitive function scores after 16 weeks of supplementation, though benefits ceased upon discontinuation. Other small trials suggest potential benefits for reducing symptoms of depression and anxiety, likely mediated by its neurogenic and anti-inflammatory effects.
Dosing Protocol
Typical dosing ranges from 1,000 to 3,000 mg per day of fruiting body extract, often divided into two or three doses. For cognitive benefits, extracts standardized to hericenones and erinacines are preferred. Mycelium extracts are often dosed lower (e.g., 250–500 mg) if highly concentrated for erinacines. Best taken with food to enhance absorption.
Safety & Contraindications
Generally well-tolerated with a high safety profile. Mild gastrointestinal discomfort or nausea can occur in some individuals. As a mushroom product, it is contraindicated in individuals with mushroom allergies. Its immunomodulatory effects suggest caution for those with autoimmune conditions or those taking immunosuppressive medications.
