Oleuropein is a prominent phenolic compound found in olive leaves and fruit, recognized for its potent antioxidant and anti-inflammatory properties. Mechanism of Action
Oleuropein, particularly in its aglycone form, is a potent inducer of autophagy. It activates the AMPK signaling pathway, which subsequently inhibits mTOR, a central regulator of cellular metabolism and growth. However, there are no published human trials explicitly evaluating oleuropein for lifespan extension or direct longevity outcomes.
Dosing Protocol
100–150 mg of oleuropein daily is the most studied range in human clinical trials, typically delivered via standardized olive leaf extract. Doses up to 1,000 mg daily have been used safely in short-term studies. It is generally taken with meals to enhance absorption and minimize gastrointestinal discomfort.
Safety & Contraindications
Oleuropein is generally well-tolerated with a high safety profile. Mild adverse effects may include gastrointestinal upset or headaches at high doses. Due to its blood pressure-lowering and hypoglycemic effects, it may interact with antihypertensive and antidiabetic medications, requiring caution and monitoring. It should be avoided during pregnancy and breastfeeding due to insufficient safety data.
