Phosphatidylserine — The Neuronal Membrane Phospholipid
Phosphatidylserine (PS) is a phospholipid that constitutes ~15% of total brain lipids and is essential for neuronal membrane integrity, signal transduction, and neurotransmitter release. It is one of the few supplements with an FDA-qualified health claim for cognitive decline reduction.
Mechanism of Action
PS maintains neuronal membrane fluidity and curvature, enabling efficient receptor signaling. It activates protein kinase C (PKC) and supports acetylcholine synthesis. PS also modulates the HPA axis, reducing cortisol response to stress. With aging, brain PS content declines, impairing neuronal function.
Human Trial Evidence
A 2010 Journal of Clinical Biochemistry and Nutrition RCT showed 300 mg/day PS improved memory and cognitive function in elderly subjects with memory complaints over 6 months. Multiple trials confirm PS reduces cortisol response to exercise stress. The FDA allows a qualified health claim: 'PS may reduce the risk of dementia and cognitive dysfunction in the elderly.'
Dosing Protocol
100–300 mg/day. 300 mg/day is the therapeutic dose used in clinical trials. Best taken with meals (fat-soluble). Soy-derived PS is the most studied; sunflower-derived PS is available for soy-free individuals. Effects typically seen after 4–8 weeks.
Safety & Contraindications
Excellent safety profile. Well-tolerated up to 600 mg/day in trials. Mild GI effects at higher doses. May mildly thin blood — caution with anticoagulants. Safe for long-term use. Soy-derived PS is safe for most soy-sensitive individuals (protein removed in processing).
