Phycocyanin (spirulina) — The Antioxidant Phycobiliprotein
Phycocyanin is a pigment-protein complex from the light-harvesting phycobilisome family, primarily found in spirulina. It exhibits potent antioxidant, anti-inflammatory, and immunomodulatory properties, making it relevant for mitigating age-related oxidative stress and chronic low-grade inflammation.
Mechanism of Action
Phycocyanin scavenges reactive oxygen species (ROS) and peroxynitrite, directly reducing oxidative damage to lipids and DNA. It inhibits key pro-inflammatory enzymes, including cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS), thereby downregulating the NF-κB signaling pathway. Additionally, it may enhance the production of endogenous antioxidants like superoxide dismutase (SOD) and catalase. Its structural similarity to bilirubin also suggests it may mimic bilirubin's physiological antioxidant effects.
Human Trial Evidence
Human trials specifically isolating phycocyanin for longevity are lacking, though numerous clinical studies have evaluated whole spirulina supplementation. These trials demonstrate improvements in lipid profiles, reduction in blood pressure, and decreased markers of oxidative stress in populations with metabolic syndrome. Direct evidence for lifespan extension in humans does not exist.
Dosing Protocol
As an isolated extract, doses of 100–500 mg/day are commonly used. When consumed as whole spirulina powder, typical doses range from 1 to 8 grams per day. It is generally taken with meals.
Safety & Contraindications
Phycocyanin and spirulina are generally recognized as safe (GRAS). Mild gastrointestinal discomfort may occur at high doses. The primary safety concern relates to the source, as contamination with heavy metals or microcystins can occur if grown in uncontrolled environments. It should be avoided by individuals with phenylketonuria (PKU) and those with autoimmune diseases should exercise caution.
