Pomegranate punicalagin — The Urolithin A Precursor
Pomegranate punicalagin is a potent polyphenol and ellagitannin that serves as a primary dietary precursor to Urolithin A. It supports longevity by promoting mitophagy, reducing oxidative stress, and modulating inflammation through its downstream gut metabolites.
Mechanism of Action
Punicalagin is a large-molecular-weight ellagitannin found abundantly in pomegranate. It is not absorbed intact but is hydrolyzed in the gut to ellagic acid, which is subsequently metabolized by the gut microbiome into urolithins, most notably Urolithin A. Urolithin A induces mitophagy, clearing damaged mitochondria and improving cellular energy capacity. It also exhibits potent antioxidant and anti-inflammatory effects by modulating pathways such as NF-κB.
Human Trial Evidence
Human trials have primarily investigated punicalagin-rich pomegranate extracts for cardiovascular health, prostate cancer, and skin microbiome benefits, showing improvements in lipid profiles and oxidative stress markers. However, there are no published human longevity trials specifically evaluating isolated punicalagin for lifespan extension.
Dosing Protocol
250–1,000 mg/day of standardized pomegranate extract (often yielding 30–300 mg of punicalagins) is the most studied range in human trials. It is typically taken orally, once daily. Optimal dosing for longevity specifically is unestablished.
Safety & Contraindications
Pomegranate extracts and punicalagins are generally well-tolerated with a high safety profile in standard doses. High doses may cause mild gastrointestinal distress, such as diarrhea. Caution is advised for individuals on medications metabolized by CYP450 enzymes, as pomegranate can inhibit certain isoforms, potentially altering drug pharmacokinetics.
