PQQ (Pyrroloquinoline Quinone) — The Mitochondrial Biogenesis Factor

Pyrroloquinoline quinone (PQQ) is a redox cofactor found in soil bacteria and trace amounts in human breast milk and certain foods (fermented soybeans, green peppers, kiwi). It is the only known dietary compound that stimulates mitochondrial biogenesis (the creation of new mitochondria) in mammals, making it a unique tool for metabolic and cognitive aging.

Mechanism of Action

PQQ activates PGC-1α (the master regulator of mitochondrial biogenesis) via CREB phosphorylation, driving the creation of new mitochondria. It also activates DJ-1 (a Parkinson's-protective protein), reduces mitochondrial ROS, and has neuroprotective effects via NMDA receptor modulation. PQQ is a potent redox cycling antioxidant.

Human Trial Evidence

A 2012 Journal of Nutritional Biochemistry RCT showed 20 mg/day PQQ significantly improved cognitive function (attention, memory) in middle-aged and elderly subjects over 12 weeks. A 2016 Functional Foods in Health and Disease study showed PQQ + CoQ10 improved subjective energy and cognitive performance. Human mitochondrial biogenesis data is primarily from animal studies.

Dosing Protocol

10–20 mg/day. Best taken with food. Often combined with CoQ10 (synergistic for mitochondrial function). Effects on energy and cognition typically noticed after 4–8 weeks. Disodium PQQ salt is the most studied form.

Safety & Contraindications

Well-tolerated at 20 mg/day in human trials. No serious adverse events reported. May cause mild headache or GI discomfort at initiation. Theoretical concern: PQQ's redox activity could theoretically promote oxidative stress at very high doses — not observed in trials. No significant drug interactions.
