Reishi extract — The Mushroom of Immortality

Reishi extract, derived from the medicinal mushroom Ganoderma lucidum, is a traditional tonic renowned for its immunomodulatory and anti-inflammatory properties. In the context of longevity, it is investigated for its ability to activate AMPK, inhibit mTOR, and reduce systemic inflammation, potentially extending healthspan.

Mechanism of Action

Reishi extract contains bioactive polysaccharides and triterpenes (such as ganoderic acids) that modulate key longevity pathways. It activates AMPK, which subsequently inhibits mTORC1 and mTORC2 signaling, promoting cellular homeostasis and autophagy. Additionally, Reishi extract downregulates the NF-κB pathway, reducing chronic inflammation and oxidative stress. It has also been shown to enhance the activity of antioxidant enzymes like glutathione peroxidase (GPX) and may exert α-Klotho-associated anti-aging effects.

Human Trial Evidence

No published human longevity trials. Animal/in-vitro evidence only. Some clinical trials have investigated its effects on immune function and metabolic parameters, but robust human lifespan data is lacking.

Dosing Protocol

1.5–9 g/day of dried mushroom or 1–1.5 g/day of concentrated extract is the most common range in clinical studies and traditional use. Standardisation to polysaccharides and triterpenes is recommended. Best taken with food. Unestablished dose-finding for longevity in humans.

Safety & Contraindications

Generally well-tolerated in healthy individuals for up to one year of oral use. Potential adverse effects include mild gastrointestinal upset, dry mouth, and dizziness. It may interact with anticoagulants, antiplatelet drugs, and immunosuppressants. Individuals with bleeding disorders or those scheduled for surgery should avoid use.
