Saffron is a spice derived from the flower of Crocus sativus, rich in bioactive carotenoids like crocin and crocetin. In the context of longevity, it is primarily studied for its neuroprotective, antidepressant, and anti-inflammatory properties, which may help preserve cognitive function and emotional well-being during aging.
Mechanism of Action
The primary active compounds in saffron, crocin and safranal, exert neuroprotective effects by modulating neurotransmitter reuptake, particularly inhibiting the reuptake of serotonin, dopamine, and norepinephrine. They also demonstrate potent antioxidant activity by neutralizing reactive oxygen species (ROS) and upregulating endogenous antioxidant enzymes like superoxide dismutase (SOD). Furthermore, saffron extracts inhibit neuroinflammation by downregulating the NF-κB pathway and reducing the expression of pro-inflammatory cytokines such as TNF-α and IL-6. Multiple randomized controlled trials have demonstrated saffron's efficacy in treating mild-to-moderate depression, often showing comparable results to standard SSRIs like fluoxetine. In cognitive health, a 22-week double-blind trial found saffron extract to be as effective as donepezil in treating mild-to-moderate Alzheimer's disease. However, specific long-term human trials evaluating saffron's direct impact on maximum lifespan or broader longevity metrics are currently lacking.
Dosing Protocol
Standard dosing in clinical trials for mood enhancement and cognitive support is typically 30 mg per day of a standardized saffron extract, often divided into two 15 mg doses. Extracts are usually standardized to contain specific percentages of crocin and safranal. It is best taken with meals.
Safety & Contraindications
Saffron is generally well-tolerated at standard doses (up to 30 mg/day). Mild adverse effects can include nausea, dry mouth, and headache. High doses (above 5 grams) can be toxic. It should be used with caution in individuals taking blood pressure medications or blood thinners due to potential additive effects, and is contraindicated in high doses during pregnancy as it may stimulate uterine contractions.
