Tributyrin SR — The Sustained-Release Butyrate Prodrug
Tributyrin is a triglyceride composed of three butyrate molecules attached to a glycerol backbone, designed to deliver butyrate directly to the lower intestine. By acting as a stable prodrug, it supports gut barrier integrity, modulates systemic inflammation, and provides a critical energy source for colonocytes, making it a key intervention for microbiome-mediated longevity.
Mechanism of Action
Tributyrin bypasses early digestion and is cleaved by pancreatic lipases in the lower gastrointestinal tract to release free butyrate. As a short-chain fatty acid, butyrate serves as the primary metabolic fuel for colonocytes and acts as a potent histone deacetylase (HDAC) inhibitor. This HDAC inhibition facilitates epigenetic modifications that downregulate the NF-κB pathway, thereby reducing the expression of pro-inflammatory cytokines. Furthermore, butyrate enhances intestinal barrier function by upregulating tight junction proteins and promotes immune tolerance by inducing regulatory T cells (Tregs).
Human Trial Evidence
No published human longevity trials. Early-stage human studies have primarily focused on safety, pharmacokinetics, and its feasibility in conditions like Parkinson's disease and cancer. While these pilot trials confirm that oral tributyrin is well-tolerated and effectively delivers butyrate to the gut, robust clinical data demonstrating lifespan or healthspan extension in healthy humans is currently lacking.
Dosing Protocol
Typical supplemental dosing ranges from 300 to 1,000 mg per day, usually taken with meals. Clinical trials investigating therapeutic applications have used significantly higher doses, but dose-finding for longevity and general healthspan remains unestablished in humans.
Safety & Contraindications
Generally well-tolerated at standard supplemental doses. The most common adverse effects are gastrointestinal, including bloating, gas, abdominal cramping, and diarrhea, particularly when initiating supplementation or at high doses. There are no well-documented severe drug interactions, but caution is advised for pregnant or breastfeeding women due to a lack of safety data.
