Vinpocetine — The Cerebral Blood Flow Enhancer
Vinpocetine is a synthetic derivative of the periwinkle plant alkaloid vincamine, traditionally used to enhance cerebral blood flow and neuroprotection. In the context of longevity, it is investigated for its potential to preserve cognitive function, mitigate neuroinflammation, and protect against age-related cerebrovascular decline.
Mechanism of Action
Vinpocetine is a synthetic derivative of the vinca alkaloid vincamine that primarily acts by enhancing cerebral blood flow and metabolism. It selectively inhibits voltage-sensitive sodium (Na+) channels, which stabilizes neuronal membranes and reduces excessive neurotransmitter release, protecting against excitotoxicity. Additionally, it acts as an inhibitor of phosphodiesterase type 1 (PDE1), leading to increased intracellular cAMP and cGMP levels, which promotes vasodilation and improves oxygen and glucose utilization in the brain. It also exhibits anti-inflammatory properties by suppressing IKK and NF-κB pathways.
Human Trial Evidence
Vinpocetine has been studied in several small human trials for cognitive impairment, dementia, and stroke recovery. A Cochrane systematic review found the evidence for its efficacy in dementia to be inconclusive, noting that while some early studies showed modest benefits in memory and concentration, the data does not strongly support clinical use for preventing or treating cognitive decline. Most human evidence is limited by small sample sizes and short durations.
Dosing Protocol
5–30 mg taken up to 3 times daily (typically 15–60 mg/day total) is the most studied range in humans. Due to its short half-life (1–2 hours), it is usually dosed in divided intervals. Best taken with food to enhance absorption.
Safety & Contraindications
Generally well-tolerated in short-term studies, but can cause gastrointestinal discomfort, nausea, sleep disturbances, headaches, dizziness, and facial flushing. It may have mild blood-thinning effects, so it should be used with caution in individuals with bleeding disorders or those taking anticoagulant medications. The FDA has issued warnings that vinpocetine consumption is associated with adverse reproductive effects, making it strictly contraindicated in women who are pregnant or could become pregnant.
