Vitamin K2 MK-7 — The Vascular Calcification Inhibitor
Vitamin K2 MK-7 is a highly bioavailable menaquinone that plays a critical role in calcium homeostasis. By directing calcium into bone tissue and away from arterial walls, it is a key intervention for preserving cardiovascular elasticity and skeletal integrity during aging.
Mechanism of Action
Vitamin K2 MK-7 acts as an essential cofactor for the enzyme gamma-glutamyl carboxylase. This enzyme catalyzes the carboxylation of specific glutamate residues in vitamin K-dependent proteins, notably osteocalcin and matrix Gla protein (MGP). Carboxylated osteocalcin binds calcium to the hydroxyapatite matrix in bone, enhancing bone mineral density. Concurrently, carboxylated MGP acts as a potent local inhibitor of vascular calcification in blood vessels. The MK-7 isoform, derived primarily from bacterial fermentation, has a significantly longer half-life in circulation compared to the MK-4 isoform, allowing for sustained systemic activation of these target proteins.
Human Trial Evidence
Observational data, such as the Rotterdam Study, demonstrate a strong inverse association between dietary menaquinone intake and severe aortic calcification, cardiovascular mortality, and all-cause mortality. In interventional trials, a 3-year double-blind, randomized controlled trial by Knapen et al. (2015) showed that 180 mcg/day of MK-7 significantly decreased arterial stiffness in healthy postmenopausal women. Additional trials have confirmed its efficacy in maintaining bone mineral density and reducing the age-related decline in bone strength.
Dosing Protocol
90–360 mcg/day is the standard evidence-based range for cardiovascular and bone health. As a fat-soluble vitamin, it should be taken orally with a fat-containing meal to maximize absorption. The 180 mcg/day dose is most commonly cited in long-term clinical trials for improving arterial stiffness.
Safety & Contraindications
Vitamin K2 is generally recognized as safe with no established upper tolerable limit. However, it strongly antagonizes the effects of vitamin K antagonist anticoagulants, such as warfarin (Coumadin); patients on these medications must strictly avoid supplementation unless directed by a physician. No significant adverse effects are reported in healthy populations at standard doses.
